About the substance
Semax is a synthetic peptide consisting of seven amino acids. Its structure combines the ACTH(4–7) fragment with Pro-Gly-Pro, often written ACTH(4–7)-Pro-Gly-Pro. This describes its molecular design, not a mixture of ACTH and separate growth factors. Semax is discussed in neurobiology because experiments have examined how it influences signals associated with nerve cells. Understanding those signals helps separate the identity of the substance from claims about a finished product.
How it works
BDNF and NGF belong to a family of signalling proteins called neurotrophins. BDNF stands for brain-derived neurotrophic factor; NGF means nerve growth factor. These proteins participate in the development, maintenance and adaptation of nerve cells. Receptors on cells receive signals and pass them into the cell. Gene expression describes how cells use genetic instructions to make molecules. Semax studies have examined changes in this system; the pen itself contains Semax, not BDNF or NGF. A change in gene activity is one biological measurement, not a direct measurement of memory or focus.
What studies have examined
Agapova and colleagues (2007) observed region-dependent changes in BDNF and NGF gene expression in rat brains after Semax administration. Dmitrieva and colleagues (2010) examined rats with experimentally reduced cerebral blood flow and reported changes in neurotrophin and receptor gene expression. The studies involved different biological conditions; they did not test this commercial pen.
How to interpret the findings
The findings help explain why Semax is discussed in relation to nerve-cell signalling and responses to injury. They do not establish that this pen improves attention, learning or everyday mental performance, or treats ADHD or neurological disease. Brain region, experimental model, formulation and delivery method affect how results should be interpreted. A study using a different preparation cannot establish how this pen behaves. Evidence for a biological mechanism, evidence for a clinical benefit and verification of a particular batch answer three different questions.
